Although it looks this way, they are not going on a vacation.

For one point, please respond and identify the base car, the coachbuilder and the year.

The coachbuilder is Miesen and the trailer is an ambulance trailer, but it isn't a Mercedes-Benz.
Locked for you until your next answer to find out the car and the year.

Yes, have a guess about the model. I reread my source and I'm not sure about the year anymore, so forget about that.

Probably 1936, yes, but I'm not sure.

One last guess about the model?

The Miesen site just describes it as a "19360000%20miesen%20operationsanh_nger%20mit%20zugfahrzeug". i.e. a 1936 Miesen Operationsanhänger mit Zugfahrzeug.  I read an inscription written diagonally on the grille beginning with a letter looking like S.

Follow that lead, although the first letter is not a S, and have a look at the unusual proportions.

It is not a Kadett.

I understand that I can risk an answer.  My first feeling was that it was not an ambulance (Krankenwagen) but an operation trailer (Operationsanhänger)

If it's not a Kadett, f the first letter is not an S (nor Six, nor SuperSix), and if I have to consider the size of the car (nor Olympia), my guess is : Regent ?

Yes, it is unlocked.

No, I don't think it is a Regent. As far as I know, at that time there was only one Opel which had its model name in chrome letters attached to the radiator.

Yes! That is the only possible explanation for the strange proportions, the chrome letters on the radiator and the fact that the trailer was probably very heavy so that at least an Olympia wouldn't be the right tow car.

For comparison, here is a Blitz radiator with those chrome letters:

That's the first time I see a Blitz looking like a saloon.  At first I was thinking of an obscure german make.  Could it be ?

This one was new to me, too. But I remember having seen some pictures of Opel Blitz buses looking like limousines, just like this one:
